Among the infectious diseases observed in bivalve molluscs, viral infections are often not well known because of a certain inadequacy of the diagnostic techniques usually applied during instances of mortality. In fact, photonic microscopy was still the base method for analysing specimens in several laboratories working on pathologies in molluscs. In 1972, Farley reported the presence of herpes-type viral particles in adult American oysters (Crassostrea virginica) for the first time. Since then, this pathogen has been detected in various marine bivalve species, usually associated with massive mortality events. Cytogenetic abnormalities associated with viral infections, including from viruses of the Herpesvirales order, have been reported in vertebrate species. Ostreid herpesvirus 1 (OsHV-1) has been detected worldwide during mortality outbreaks of the Pacific oyster Crassostrea gigas. On the other hand, a high proportion of aneuploid cells in somatic tissues have been observed in C. gigas. In this study, we analysed the putative association between aneuploidy levels and the detection of OsHV-1 in gills of C. gigas, the Portuguese oyster C. angulata and their F1 hybrids cultured in Ria Formosa (Portugal). As the marine mussel Mytilus edulis is commonly used as a sentinel species, it would be useful to develop a primary culture of the target organs most often in contact with the marine environment. This study reports an improved method for dissociating the digestive gland and gills of M. edulis and considers the effect of mussel storage on cell viability and functionality before culture initiation. Viability and enzymatic activities such as those of esterase and peroxidase were monitored by flow cytometry, a sensitive, objective technique allowing large volumes of cells to be counted within a short time.
